RIPA Lysis Buffer Recipe

RIPA lysis buffer (short for radioimmunoprecipitation assay  buffer) is commonly used to cell lysis prior to immunoprecipitation. It is very useful for the extraction of cellular proteins, but is not appropriate for co-immunoprecipitation studies, as the SDS may disrupt many protein-protein interactions.

RIPA Lysis Buffer is usually composed as follows:

  • 1% NP-40
  • 0.1% SDS
  • 50 mM Tris-HCl pH 7.4
  • 150 mM NaCl
  • 0.5% Sodium Deoxycholate
  • 1 mM EDTA

Recipe for 10x RIPA lysis buffer (250 ml):

  • 25 ml NP-40 detergent.
  • 12.5 ml 20% SDS
  • 125 ml of 1M Tris-HCl pH 7.4
  • 75 ml 5M NaCl
  • 12.5 g Sodium Deoxycholate
  • 5 ml 0.5M EDTA
  • 7.5 ml water
